Sodium cholesteryl sulfate
Sodium cholesteryl sulfate is an endogenous sterol sulfate and a component of cell membranes. It is an orally available, NPC2-targeted cholesterol biosynthesis agonist that activates SREBP2 by competitively binding to NPC2, promoting cholesterol synthesis (EC₅₀ = 50 μM). The compound exhibits anti-inflammatory properties, alleviating ulcerative colitis in mouse models by repairing the intestinal mucosal barrier and inhibiting inflammation through regulation of serine protease activity and PKCη signaling. It also enhances the self-assembly ability of mitoxantrone hydrochloride for antitumor activity via nano-delivery systems. Additionally, sodium cholesteryl sulfate supports platelet adhesion, regulates selective protein kinase C isoforms, and modulates PI3K specificity in keratinocyte differentiation. As a pharmaceutical excipient, it functions as an emulsifier, surfactant, and stabilizer in lipid-based formulations, helping to form and stabilize emulsions, liposomes, and colloidal drug delivery systems.
