Description	DSG-PEG 2000 is a non-ionic PEGylated lipid widely used as a steric-stabilizing excipient in lipid-based formulations. It consists of a hydrophobic distearoyl-rac-glycerol anchor covalently linked to a hydrophilic polyethylene glycol (PEG 2000) chain, enabling the DSG moiety to embed in the lipid bilayer while the PEG segment extends outward to form a hydrated, neutral surface layer. This structure minimizes non-specific interactions, enhances colloidal stability, and supports prolonged circulation. DSG-PEG 2000 is used in lipid nanoparticle (LNP) and liposome preparation for siRNA, mRNA, DNA, and small molecule delivery. Notably, DSG-PEG dissociates more slowly from LNPs than DMG-PEG due to its longer C18 alkyl chains, resulting in longer circulation times. It has been employed in GalNAc-LNP formulations for CRISPR base editing delivery, where the DSG anchor showed considerably higher potency than cholesterol or C20 anchors.
