mPEG-ALD (mPEG-Aldehyde) will undergo reductive amination with primary amines in the presence of sodium borohydride. The aldehyde group is different from other electrophilic active groups in that it only reacts with the amine group. Although the reactivity of the aldehyde group is lower than that of the NHS active ester, its reaction conditions are mild (pH 6-9.5, 6-24 hours), and it is easy to connect PEG to the surface of proteins or other materials. At lower pH, mPEG-ALD will selectively react to the N-terminus of the protein. The stability of its connection (the formation of secondary amines through reduction reactions) is extremely important for the preparation of immobilized enzymes. The protein modified in this way can retain the chargeability of the amino group and the protein in the solvent, which is very important for maintaining the structure and activity of the protein. The quantitative analysis of lysine in protein hydrolysates by amino acid analysis method can easily characterize these coupling products. mPEG-ALD is a general reagent for N-terminal PEGylation of proteins, and its purity is about 80%.
